The first recorded lord of the manor was Cospatric de Samlesbury, and it is believed that he lived in the Lower Hall on the banks of the river Ribble. During the 13th Century, the manor was divided between the two granddaughters, Elizabeth and Cecily de Samlesbury as there were no male heirs. Cecily went on to marry John Deuyas , having a daughter Alicia, who was married to Gilbert de Southworth around the year 1325. The Deuyas coat of arms was adopted by Gilbert who was responsible for building the Great Hall. Their descendants kept hold of the manor until 1677/78, at which point is was sold to the Bradylls, who rented out the Hall to handloom weavers, before it was converted into the Bradyll Arms.

Joseph Harrison eventually bought the estate, after it passed through the hands of Thomas Cooper, and he spent a large amount of money restoring Samlesbury Hall before it was left to ruin. Finally a group of six businessmen bought Samlesbury Hall in 1924, saving it from being demolished by a building firm who planned to erect a housing estate. The Samlesbury Hall Trust was started in 1925 at has been administered by The Trust ever since.
